Induction of Specific Humoral Immune Response in Mice against a Pseudomonas aeruginosa Chimeric PilQ/PilA Protein

چکیده مقاله:
Background: Pseudomonas aeruginosa, an opportunistic pathogen, is a common cause of healthcare-associated infections in immunocompromised individuals. The rapid emergence of multidrug-resistant strains has made P. aeruginosa infections progressively difficult to treat. In this study we evaluated the effect of a chimeric protein containing a P. aeruginosa PilQ fragment and the PilA disulfide loop (PilA-DSL) on the humoral immune response in BALB/c mice. Methods: A chimeric gene encoding an immunogenic region of PilQ and the PilA-DSL was synthesized. Following bacterial expression and purification, the protein was administered to mice and the humoral immune response analyzed. The resulting antibodies were analyzed using an opsonophagocytic killing assay. Results: The anti-recombinant protein antibody titer was significantly greater in immunized mice than in controls. In addition, antibody titers were significantly increased after booster immunizations, and the immunizations induced opsonophagocytosis of P. aeruginosa PAO1. Conclusions: These results suggest that an anti-adhesion-based vaccination may be effective in preventing P. aeruginosa infections. Further studies are needed to evaluate the abilities of such bivalent proteins to induce strong immune responses.

Assessment of humoral immune response of a Cytomegalovirus DNA-vaccine candidate in BALB/c mice
Introduction: Glycoprotein B (gB) is the major antigen for induction of humoral responses against human cytomegalovirus (HCMV) making it an attractive candidate for immune prophylaxis. In the present study, the humoral immune response of BALB/c mice to a truncated HCMV gB protein fused with GFP was evaluated. Methods: The truncated gB coding sequence was synthesized and cloned in pEGFPN1 eukary...
متن کاملPRODUCTION AND CHARACTERIZATION OF HUMORAL IMMUNE RESPONSE AGAINST MUSTARD GAS
One of the promising aspects of the immunological research on chemical war gas is to investigate the immunogenicity of some hazardous compounds such as mustard gas. Mustard gas is categorized as a "hapten" based on its physical and chemical properties. Haptenic chemicals which do not possess immunogenicity could be immunogenic experimentally when conjugated with a suitable protein carrier...

conclusions our results demonstrated that r-fla-a could induce cellular and humoral immune response as proper stimulant of poly-isotypic humoral responses. results immunized mice with adjuvanted flagellin showed a considerably increased lymphocyte proliferation compared with the control group (p = 0.004). high level of il-4 and ifn-γ secretion was observed in immunized mice compared with the co...
متن کاملInduction of humoral immune responses and inhibition of metastasis in mice by a VEGF peptide-based vaccine
Objective(s): Blocking of vascular endothelial growth factor (VEGF) plays a pivotal role in inhibition of metastasis and is a target for development of anti-angiogenic agents. In this study, a peptide-based vaccine was designed and its potential for induction of humoral immune responses, generation of neutralizing antibodies, inhibition of tumor growth and metastasis w...

Background: DNA vaccines are third generation vaccines which have made promises to combat infectious diseases. Cationic liposomes are used as effective delivery systems for DNA vaccines to generate stronger immunity. Objective: Encapsulation of pcDNA3.1+PA plasmid, encoding protective antigen (PA) of Bacillus anthracis (B. anthracis) into cationic liposomes, and evaluation of its effect on spec...
متن کاملimmune priming against Drosophila Pseudomonas aeruginosa is short - lasting and depends on cellular and humoral immunity
Immune responses are traditionally divided into the innate and the adaptive arm, both of which are present in vertebrates, while only the innate arm is found in invertebrates. Immune priming experiments in and Drosophila melanogaster other invertebrates during the last decade have challenged this dogma, questioning the boundaries between innate and adaptive immunity. Studies on repeated inocula...
